Transcription factor 19 (TCF19) is a DNA-binding transcription factor that plays a crucial role in the regulation of genes involved in the cell cycle, particularly at the G1/S transition. It contains a forkhead association (FHA) domain and a PHD-type zinc finger region, which are implicated in chromatin interaction and transcriptional regulation. TCF19 is necessary for cell proliferation and survival, particularly in pancreatic β-cells, where it is linked to cell cycle progression, the maintenance of ER homeostasis, and apoptosis regulation. Overexpression or functional variants of TCF19 have been associated with various cancers, such as thyroid, hepatic, and colorectal cancers, often acting as an oncogenic factor by promoting tumor cell proliferation and immune evasion. Genetic variants within TCF19 are associated with increased risk for autoimmune diseases, type 1 diabetes, and thyroid cancer, making TCF19 an emerging biomarker and a potential therapeutic target in oncology and metabolic diseases.
