Overview

Transcription factor AP-4 (TFAP4) is a member of the basic helix-loop-helix (bHLH) transcription factors encoded by the TFAP4 gene in humans[1][3]. It binds the E-box DNA sequence (CAGCTG) in promoters to regulate target genes, acting as both activator and repressor depending on context[1][3]. TFAP4 is a direct target of c-MYC and plays a central role in promoting cell cycle progression and proliferation, in part by repressing cell cycle inhibitors such as p21, and ensuring the fidelity of mitosis, with loss of regulation leading to DNA damage and mitotic defects[6]. In T-cell development, TFAP4 represses CD4 gene expression in immature thymocytes, cooperating with Runx proteins and interacting with histone deacetylases to modulate chromatin structure and gene accessibility[2]. Elevated TFAP4 expression is associated with aggressive tumor phenotypes and poor prognosis; it can directly activate the Wnt/β-catenin signaling pathway by transcriptionally regulating downstream targets such as DVL1 and LEF1, thereby increasing expression of critical cell proliferation genes and promoting tumorigenesis[5][6].
