Transcription factor CP2-like 1 (TFCP2L1) is a member of the CP2 subfamily within the LSF/Grainyhead family of transcription factors[1][3]. It is critical for maintaining pluripotency and self-renewal in embryonic stem cells, primarily via direct regulation of target genes such as ESRRB and KLF4, and is necessary for normal development of tissues like the kidney and salivary gland[3][4][5]. In adult tissues, TFCP2L1 coordinates the development of intercalated and principal cells in the renal collecting ducts, participating in acid-base and salt-water homeostasis by regulating the expression of V-ATPase complex subunits and Notch signaling intermediates[1][5]. Beyond development, abnormal TFCP2L1 activity is linked to cancer stem cell maintenance and poor prognosis in cancers such as hepatocellular carcinoma, where it may promote resistance to targeted therapies, and in renal cancers where deregulation contributes to oncogenic reprogramming and tumor aggressiveness[2][3].
