Transcription factor E3 (TFE3) is a bHLH-Zip transcription factor belonging to the MiTF/TFE family. It regulates gene expression by binding to E-box and CLEAR-box sequences, playing a key role in lysosomal biogenesis, autophagy, immune response, and cellular metabolism. TFE3 activity is regulated by mTOR-dependent phosphorylation, controlling its nuclear translocation. Chromosomal translocations involving TFE3 are implicated in renal cell carcinomas and alveolar soft part sarcoma. It also interacts with Gα16, influencing cardiac hypertrophy. TFE3 is a therapeutic target in cancer.
