Overview

Transcription factor E47 is a basic helix-loop-helix (bHLH) protein that binds specific DNA sequences to regulate gene expression central to lymphocyte development[1][2][4]. E47 forms homodimers and heterodimers, directly activating or repressing key genes critical for the differentiation and maturation of B and T cells[1][4]. It binds E-box motifs (DNA sequence -CACCTG-) in enhancer regions to modulate the transcription of genes such as immunoglobulin heavy-chain, kappa light-chain, RAG-1, RAG-2, Oct-2, and CD5[1][5]. E47 regulates cell cycle checkpoints, promotes cell cycle arrest by repressing Cdk6 and other cell cycle regulators, and modulates genes involved in cell survival (Bid, Bim, caspase-3/6)[4]. Its protein structure consists of a bHLH domain that dimerizes and specifically recognizes DNA, with additional regions affecting dimerization and regulatory capacity[2][6]. E47 also acts upstream of numerous transcriptional regulators governing lymphoid fate decisions (such as Hes1, Ets2, Gata3, Foxo1, Xbp1, and others)[4], and its activity is modulated by factors such as Id2 and Eto2[4]. Dysfunction or alteration in E47-binding at enhancer sites has been implicated in acceleration of lymphoid malignancies and influence on retroviral enhancer function in leukemia viruses[3].
