Transcription factor EB (TFEB) is a master regulator of cellular homeostasis and metabolism. As a member of the microphthalmia family of basic helix-loop-helix–leucine-zipper (bHLH-Zip) transcription factors (MiT family), TFEB functions as a master regulator of multiple cellular processes. It regulates lysosomal biogenesis, autophagy, mitochondrial biogenesis, and various metabolic processes. TFEB is activated by cellular stressors and has shown therapeutic potential in neurodegenerative diseases, while also playing complex roles in cancer and inflammatory responses.
