Transcription factor IIIB 50 kDa subunit (BRF2) is a core component of the RNA polymerase III transcription initiation factor complex (TFIIIB), essential for the initiation of transcription of genes with upstream promoter elements, particularly small nuclear RNA genes. BRF2 is a TFIIB-like factor that is directly recruited to the TATA box by interacting with the TATA-binding protein, and is necessary for forming a stable pre-initiation complex for RNA polymerase III. Aberrations in BRF2 expression and function have been linked to the development and progression of various cancers, making it a potential biomarker and emerging therapeutic target, especially in oncology.
Not applicable for approved drugs. Investigational inhibitors would block the transcriptional activation function of BRF2, suppressing RNA polymerase III-dependent gene transcription and reducing tumor-associated gene expression
