Overview

Transcription factor-like 5 protein (TCFL5) is a basic helix-loop-helix transcription factor that primarily acts as a DNA-binding transcriptional repressor specific for RNA polymerase II. TCFL5 is expressed in a cell-type and stage-specific manner during spermatogenesis, with key roles in regulating germ cell differentiation. Isoform diversity gives rise to distinct biological effects, notably in colorectal cancer, where different TCFL5 variants can be either pro-tumorigenic or anti-tumorigenic through modulation of pluripotency genes. Additionally, TCFL5 is implicated in chemotherapy resistance in hematologic malignancies, possibly via regulation of antioxidant mechanisms. Its precise mechanisms involve modulation of transcriptional programs related to cell cycle, proliferation, differentiation, and survival
