Overview

SRY-box transcription factor 10 (SOX10) is a nuclear transcription factor belonging to the high-mobility group (HMG)-box family. It plays a central role in the specification, differentiation, maturation, and maintenance of neural crest-derived cell lineages, including melanocytes, Schwann cells, oligodendrocytes, and other glia[1][2][4][6]. SOX10 acts as a sequence-specific DNA-binding factor, activating transcription of target genes essential for development of the peripheral and central nervous system, pigment cells, and certain epithelial tissues[4][5]. Its activity is critical for proper embryogenesis and for the normal maintenance of differentiated states postnatally. Mutations in SOX10 cause a range of neurocristopathies, including Waardenburg syndrome, Hirschsprung disease, and various forms of leukodystrophy[4][6]. In cancer biology, SOX10 is an important lineage marker and functional driver, especially in melanoma, where it supports stem cell properties and impacts invasion, metastasis, and therapy resistance[1][3]. Its expression is sensitive and specific for neural crest–derived tumor diagnosis in pathology. There are currently no approved pharmacologic agents that directly target SOX10 in the clinic, but loss- or gain-of-function in this factor has profound developmental and disease implications[1][4].
