Overview

Transcription factor SOX-6 is a member of the SOX (SRY-related HMG-box) family, characterized by a highly conserved DNA-binding HMG domain that allows it to regulate gene expression by binding to specific DNA motifs (UniProt P35712). It plays a critical role in several developmental processes, including chondrogenesis, where it cooperates with SOX5 and SOX9 to activate cartilage-specific genes, and erythropoiesis, where it facilitates the switch from fetal to adult hemoglobin (NCBI Gene 6665). In the central nervous system, SOX6 is essential for the proper development of interneurons and oligodendrocytes, and its haploinsufficiency is the primary cause of Tolchin-Le Caignec syndrome, a neurodevelopmental disorder (PubMed 32442411). Beyond development, SOX6 is implicated in metabolic and cardiovascular diseases, acting as a regulator of adipogenesis and blood pressure through its effects on vascular smooth muscle cells (PubMed 33226715). In oncology, it functions as a context-dependent regulator, often acting as a tumor suppressor in gliomas and ovarian cancer, though its dysregulation can promote progression in other malignancies (PubMed 31428241). While no small molecule drugs are currently approved, SOX6 mRNA is an active target for experimental RNA-based therapeutics, such as antisense oligonucleotides and siRNAs, designed to modulate its expression in chronic conditions like hypertension and diabetes (PubMed 33226715).
