Transcription factor Spi-B (SPIB)

Target
SPIB
Molecular classification
Transcription factor, ETS family protein
01

Overview

Transcription factor Spi-B (SPIB) is an ETS family protein acting as a DNA-binding transcriptional activator within lymphoid lineages. It is critical for the development, structural maturation, and antigen-uptake function of specialized immune cells called M cells in the gut, and plays a key role in B cell differentiation[1][3]. In cancer, SPIB can activate key signaling pathways (NFkB, JNK) as part of its tumor suppressor activity[5]. Dysfunction or deficiency of Spi-B affects immune responses, host-microbe interactions, and tumor suppression, making it a molecular target of interest in immunology and oncology[1][5].

Biological functions

Regulation of gene expressionLymphocyte differentiation and maturation (notably in B cells and M cells)Immune system responseTumor suppression via activation of NFkB/JNK signaling

Disease associations

Cancer (e.g., colorectal cancer tumor suppression)Infection (regulation of M-cell differentiation and pathogen uptake)Immune system disorders (role in lymphocyte and immune cell function)
04

Safety considerations

Since Spi-B is highly lineage-specific and regulates immune cell differentiation, systemic targeting could present risks in immune function or cause off-target effects related to immune homeostasis
