Overview

Transcription initiation factor TFIID subunit 12 (TAF12) is a vital component of the TFIID multiprotein complex, which serves as the primary scaffold for the assembly of the RNA polymerase II preinitiation complex on gene promoters (UniProt Q16514). TAF12 is characterized by a histone-fold domain that facilitates its heterodimerization with TAF4, a structural interaction necessary for the integrity of the TFIID core (PubMed: 25135911). This subunit plays a specialized role in regulating the transcription of genes involved in cell growth, differentiation, and the stress response (PubMed: 30249654). In oncology, TAF12 has been identified as a key dependency in several cancers, most notably in acute myeloid leukemia (AML), where it is required to maintain the oncogenic transcriptional program (PubMed: 26373484). Although there are currently no FDA-approved drugs that directly target TAF12, it is considered an attractive target for the development of small-molecule inhibitors or proteolysis-targeting chimeras (PROTACs) in transcription-addicted tumors. However, the therapeutic window for targeting TAF12 may be narrow because of its fundamental requirement in basal transcription across all healthy tissues, posing significant safety concerns regarding systemic toxicity.
