Transcription initiation factor TFIID subunit 6 (TAF6) is a protein encoded by the TAF6 gene in humans and is one of the smaller subunits of the TFIID complex, which is essential for the initiation of RNA polymerase II-dependent transcription. TFIID, composed of the TATA-binding protein (TBP) and TBP-associated factors (TAFs), binds to the core promoter to position the polymerase, serves as a scaffold for the transcription complex, and channels regulatory signals. TAF6 interacts weakly with TBP but strongly with TAF1 and helps stabilize the architecture of TFIID by connecting distinct structural modules. Alternative splicing results in several isoforms, with certain isoforms regulating apoptosis through influence on transcription and by initiating expression of genes such as GADD45A and CDKN1A/p21.
