Overview

Transcription initiation protein SPT3 homolog (SUPT3H) is a component of the highly conserved SAGA (Spt-Ada-Gcn5 acetyltransferase) and STAGA (SPT3-TAF9-GCN5 acetyltransferase) transcriptional coactivator complexes, which play important roles in regulating chromatin accessibility and transcription by RNA polymerase II[1][2][3][6]. SUPT3H contains histone-fold domains and contributes structurally to these complexes, mediating protein-protein interactions important for complex integrity. While not essential for core promoter binding or global transcription initiation in mammals, SUPT3H is required for the expression of a specific subset of genes and is important for cell growth and self-renewal in embryonic stem cells[1]. There is currently no evidence that SUPT3H is a direct therapeutic target or that drugs bind specifically to this protein. SUPT3H does not function as a classical receptor, enzyme, or transporter but rather as a transcriptional coactivator within multi-protein complexes that regulate gene expression by modifying chromatin structure[2][4][6]. Disease relevance is noted with cleidocranial dysplasia, but mechanistic links remain limited[2].
