Transcriptional adapter 2-alpha (TADA2A) is a nuclear protein that acts as a transcriptional coactivator, functioning as an adaptor to bridge sequence-specific DNA-binding transcription factors with the general transcription machinery of RNA polymerase II. It serves as an integral component of large multi-subunit histone acetyltransferase complexes, notably the PCAF and ATAC complexes, which acetylate histones H3 and H4 and facilitate chromatin remodeling and transcriptional activation. TADA2A is essential for modulating chromatin accessibility, promoting gene expression through nucleosome acetylation, and is involved in several critical cellular processes, including cell cycle progression, DNA repair, and apoptosis through its regulatory interactions. The gene is subject to alternative splicing and has several transcript variants, and while not itself a direct therapeutic drug target, its dysregulation is associated with cancer, with circTADA2A—a circular RNA derived from TADA2A—emerging as a potential biomarker for certain malignancies
