Overview

Transcriptional adaptor 3 (TADA3) is a conserved adaptor protein that acts as a coactivator for transcriptional activators, functioning as a critical subunit of histone acetyltransferase (HAT) complexes such as PCAF and ATAC, thereby regulating chromatin accessibility and gene expression[1][2][4][5]. TADA3 is required for efficient acetylation of histones, modulates cell cycle progression, and stabilizes the tumor suppressor p53, facilitating p53-mediated DNA damage response and apoptosis[1][3][5][6]. It directly interacts with diverse transcription factors, including p53, retinoic acid receptor alpha, and estrogen receptors, as well as other coactivators, and its loss or dysfunction has been implicated in oncogenic transformation, notably in HPV-related cancers and in the progression and aggressiveness of several carcinomas[3][5].
