Transcriptional enhanced associate domain (TEAD) proteins are a family of transcription factors that serve as the primary downstream effectors of the Hippo signaling pathway, which is crucial for regulating cell proliferation, tissue homeostasis, stem cell function, and organ size. TEADs require coactivators—most notably Yes-associated protein (YAP) and transcriptional co-activator with PDZ-binding motif (TAZ)—to activate gene transcription. High expression or dysregulation of TEADs is linked to various cancers and disrupting the interaction between YAP/TAZ and TEAD is an active area for anti-cancer drug development.
