Overview

Transducin beta-like protein 2 (TBL2) is a member of the beta-transducin protein family encoded by the TBL2 gene on chromosome 7q11.23[1][6]. The protein is possibly involved in intracellular signaling and has roles in the endoplasmic reticulum (ER) stress response, particularly the unfolded protein response[2]. Genetic deletion of TBL2 has been implicated in Williams-Beuren syndrome, a neurodevelopmental disorder caused by deletions at chromosome region 7q11.23[1]. TBL2 has also been identified as a potential driver gene in certain cancers, such as lung adenocarcinoma, where it may promote tumor growth by adapting to ER stress[5]. There are no established drugs reported to specifically target TBL2, and its direct therapeutic or biomarker utility remains unclear.
