Overview

Transducin-like enhancer of split 2 (TLE2) is a transcriptional corepressor protein that belongs to the Groucho/TLE family of WD repeat domain-containing proteins[3]. It interacts with various DNA-binding transcription factors to mediate transcriptional repression, functioning primarily in the nucleus. TLE2 is involved in the negative regulation of gene expression and canonical Wnt signaling pathways[3]. In the context of Kaposi's sarcoma-associated herpesvirus (KSHV) infection, TLE2 forms a nuclear complex with the viral replication and transcription activator (RTA), repressing RTA-driven autoactivation and viral gene transactivation, thus contributing to viral latency maintenance[1][2][3]. TLE2 exerts its corepressor function via specialized domains (Q, SP, and WDR), and its activity can antagonize transcriptional programs essential for cell fate and disease processes including cancer and viral infection[1][2][3].
