Transducin-like enhancer protein 4 (TLE4) is a member of the TLE/groucho family of transcriptional corepressors that do not directly bind DNA but regulate gene expression via interaction with transcription factors, including Hes1 and PAX5. TLE4 possesses an N-terminal Q domain for tetramerization and a C-terminal WD repeat domain for protein interaction; it acts as a critical regulator of hematopoiesis, B-cell differentiation, and inflammatory responses. Specifically, TLE4 represses the expression of pro-inflammatory cytokines such as IL-6 and IL-12 in macrophages, modulating immune homeostasis. TLE4 also contributes to the negative regulation of the canonical Wnt signaling pathway and is important for normal skeletal and bone marrow development. In disease, TLE4 deletion or dysfunction is implicated particularly in acute myeloid leukemia as a synergistic factor in oncogenesis and in immune dysregulation.
