Overview

Transducin-like enhancer protein 6 (TLE6) is a member of the Groucho/transducin-like enhancer of split family of transcriptional co-repressors, characterized as a core component of the subcortical maternal complex (SCMC) in mammalian oocytes and early embryos[1][3][6]. It consists of 581 amino acids, contains five WD40 repeat domains, and is critical for the regulation and maintenance of the actin cytoskeleton during oocyte maturation and symmetric zygotic cell division[1][2][4][6][7]. Loss of TLE6 results in failed embryogenesis beyond the two-cell stage, leading to preimplantation embryonic arrest and infertility in both animal models and humans[1][2][6][7]. TLE6 is not considered a classical therapeutic target (such as a receptor or enzyme), but pathogenic variants serve as genetic biomarkers for certain infertility syndromes[2][3][7].
