Overview

Transforming acidic coiled-coil-containing protein 1 (TACC1) is a member of the TACC protein family, characterized by a large carboxy-terminal coiled-coil domain. TACC1 is implicated in regulating transcription, translation, and centrosome dynamics by interacting with a variety of protein complexes. It functions downstream of growth factor and oncogenic signaling, particularly Ras and PI3K/AKT/mTOR pathways, promoting cell survival and proliferation. TACC1 is frequently overexpressed or aberrantly spliced in several cancers and can enhance tumor formation and decrease apoptosis. It interacts with proteins involved in cell division and the mitotic spindle, such as Aurora kinases and CKAP5, but has minimal direct association with microtubule stabilization. While TACC1 is considered a potential biomarker and cancer target, no direct therapeutics targeting TACC1 are currently approved.
