Overview

Transforming growth factor beta-1-induced transcript 1 protein (TGFB1I1) is a multi-functional adapter and coactivator protein, encoded by the TGFB1I1 gene in humans. Its expression is induced by transforming growth factor beta-1 (TGFB1) and hydrogen peroxide. It is predominantly localized at the focal adhesion complexes, but also shuttles between the cytoplasm and nucleus. TGFB1I1 functions as a scaffold at focal adhesions, coordinates protein-protein interactions, and acts as a nuclear coactivator for several hormone receptors, including androgen and glucocorticoid receptors. These activities allow TGFB1I1 to regulate signal transduction, cellular proliferation, migration, differentiation, and senescence. Its involvement in cancer (including prostate cancer) and its role as an androgen receptor coactivator highlight its therapeutic relevance[1][2][8].
