The Transforming Growth Factor beta 1 signaling pathway is a multi-protein cascade comprised of **TGF-β1 ligand** (a cytokine), its cell-surface **serine/threonine kinase receptors** (primarily TGFβRI/ALK5 and TGFβRII), and nuclear/intracellular effector proteins, most notably the **SMAD family** of transcription factors[5][6][3][4]. After ligand binding, TGF-β type II receptor recruits and phosphorylates the type I receptor, which activates SMAD2/3 via phosphorylation. Activated SMAD complexes translocate to the nucleus to regulate gene expression governing proliferation, differentiation, apoptosis, immune homeostasis, and fibrosis[3][6][5]. Dysregulation of this pathway is implicated in cancer, fibrosis, inflammation, cardiovascular disease, and more. While numerous drugs target this pathway (mainly at the ligand or receptor level), therapeutic intervention is challenged by its widespread physiological roles and the risk of significant adverse effects[9][5][6].
