Transforming growth factor beta (TGF-β) activation refers to the process by which latent TGF-β, a multifunctional cytokine, is converted into its active form capable of binding and signaling through its receptors. TGF-β plays critical roles in embryonic development, cell growth, differentiation, motility, apoptosis, immune regulation, and tissue homeostasis. Activation involves releasing mature TGF-β from its latent complex so it can bind to cell surface receptors. Several mechanisms are known: Proteolytic cleavage, Integrin-mediated activation, Thrombospondin 1 interaction and Environmental factors. Once activated: Active TGF-β binds first to type II receptor (TβRII), The ligand-receptor II complex recruits type I receptor (TβRI), Phosphorylated type I receptor propagates intracellular signals primarily via SMAD transcription factors. Dysregulation of activation leads to pathological conditions.
