The transforming growth factor beta (TGF-β) family of ligands is a large group of structurally related cytokines that play essential roles in embryonic development, adult tissue homeostasis, stem cell fate determination, immunity, and disease processes such as cancer. The mammalian TGF-β family consists of 33 members that signal through a conserved mechanism involving serine/threonine kinase receptors and SMAD transcription factors. Dysregulation of TGF-beta signaling is implicated in various diseases including cancer progression/metastasis, fibrosis, autoimmune disorders, and developmental abnormalities.
TGF-beta family ligands bind to type I and type II serine/threonine kinase receptors, activating downstream SMAD signaling pathways to regulate gene expression.
