Transforming growth factor beta (TGF-β) is a multifunctional cytokine and a member of the TGF-β superfamily. It plays essential roles in various cellular processes, including cell proliferation, differentiation, apoptosis, adhesion, motility, and immune regulation. Dysregulation of TGF‑β signaling has been implicated in several diseases, including fibrosis, cancer, and autoimmune disorders.
TGF‑β ligands bind to type II receptors (e.g., TβRII), which then recruit and phosphorylate type I receptors (e.g., ALK5/TβRI). This activated receptor complex initiates intracellular signaling cascades primarily via phosphorylation of SMAD transcription factors, leading to regulation of gene expression. Non-SMAD pathways, including MAP kinase pathways, PI3K/Akt pathway, and Rho GTPases, can also be activated.
