Overview

Transforming growth factor beta (TGF-β) pathway-related messenger RNAs (mRNAs) are the transcripts encoding the ligands, receptors, and intracellular signaling proteins of the TGF-β superfamily [1][2]. This pathway is a critical regulator of cell growth, differentiation, and immune homeostasis, but its dysregulation is a key driver in cancer progression and fibrotic diseases [3][4]. In the context of malignancy, these mRNAs are often overexpressed, leading to an immunosuppressive tumor microenvironment and promoting the epithelial-mesenchymal transition (EMT), which facilitates metastasis [4]. In fibrotic disorders, the over-expression of TGF-β pathway mRNAs results in the excessive accumulation of extracellular matrix components, leading to organ failure [5]. Therapeutic strategies targeting these mRNAs primarily involve antisense oligonucleotides (ASOs) and RNA interference (RNAi) to selectively reduce the translation of pathogenic proteins [6]. However, the dual role of TGF-β as both a tumor suppressor and a promoter, along with its essential functions in normal tissue repair, poses significant challenges for therapeutic development and safety [7].
