Overview

Transforming growth factor-beta receptor-associated protein 1 (TGFBRAP1) is a cytoplasmic protein involved in the trafficking and fusion of early endosomes as a specific subunit of the CORVET (Class C core vacuole/endosome tethering) complex. It also binds to transforming growth factor-beta (TGF-beta) receptors and modulates TGF-beta/activin signaling, particularly by facilitating interactions with SMAD4 and influencing downstream signaling events. TGFBRAP1 is not a receptor, enzyme, or classical drug target but a tethering complex protein important for vesicle dynamics in the endocytic pathway, thus playing crucial roles in intracellular transport and signal transduction by regulating membrane fusion and trafficking. Mutations or dysregulation of TGFBRAP1 are implicated in some pathologies, including Long QT syndrome and neuroblastoma, though its direct role as a disease driver is not definitively characterized. There are no known drugs that directly target TGFBRAP1, nor is it established as a therapeutic target or clinical biomarker at this time.
