Overview

The Transforming growth factor-beta (TGF-β) receptor type II signaling complex is a fundamental heterotetrameric unit responsible for transducing signals from TGF-β ligands into the cell (UniProt P38438; Wikipedia). It typically consists of two TGF-β receptor type II (TGFBR2) subunits and two TGF-β receptor type I (TGFBR1, also known as ALK5) subunits (NIH: PMC3481076). Upon ligand binding, the constitutively active kinase domain of TGFBR2 phosphorylates the GS domain of TGFBR1, thereby activating it to initiate the canonical SMAD signaling pathway and various non-canonical pathways like MAPK and PI3K/AKT (NIH: PMC4299710). This complex plays a dual role in human physiology: it acts as a potent tumor suppressor in early-stage cancers by inducing cell cycle arrest and apoptosis, but it promotes tumor progression, metastasis, and immunosuppression in advanced stages (NIH: PMC4299710). Consequently, it is a high-priority therapeutic target, with drug development focusing on small molecule kinase inhibitors, monoclonal antibodies, and ligand traps to treat oncology and fibrotic diseases (PubMed: 28246548).
