Transforming growth factor beta regulator 1 (TBRG1)

Target
TBRG1
Molecular classification
Nuclear protein, Intracellular signaling peptide/protein, Other (not a classical receptor, enzyme, transporter, ion channel, or transcription factor)
01

Overview

Transforming growth factor beta regulator 1 (TBRG1) is a nuclear growth-inhibitory protein with established tumor-suppressing activity in human cancers and key roles in cell cycle regulation. It can activate p53/TP53, induce G1 cell cycle arrest, and restrict cell proliferation in cooperation with CDKN2A, but its growth-inhibitory function does not require either p53 or CDKN2A. TBRG1 also contributes to chromosomal stability maintenance and can regulate innate immunity, as demonstrated in invertebrates. Currently, no drugs are known to directly target TBRG1 for therapeutic purposes.

Mechanism of action

Not applicable—no known drugs targeting TBRG1 directly. Its biological mechanisms include activation of p53/TP53, induction of G1 cell cycle arrest, and cooperation with CDKN2A to restrict proliferation.

Disease associations

Safety considerations

None documented for targeting TBRG1 directly; main therapeutic challenge is that loss of TBRG1 promotes tumorigenesis, highlighting its tumor suppressor functions
