The transforming growth factor beta (TGF-β) signaling pathway is a highly conserved cellular communication system that regulates a wide array of biological processes in both embryonic development and adult tissue homeostasis. It is crucial for controlling cell growth, differentiation, migration, apoptosis, immune regulation, vascular function, and maintenance of cellular homeostasis. The pathway is initiated by TGF-β ligands binding to type II and type I receptor serine/threonine kinases, leading to SMAD protein phosphorylation and translocation to the nucleus to regulate gene transcription, as well as activation of non-SMAD signaling pathways. Dysregulation of this pathway is implicated in various diseases, including cancer, fibrosis, and autoimmune disorders.
