Transforming growth factor beta type II receptor (TGFBR2) is a transmembrane serine/threonine protein kinase that plays a central role in the TGF-β signaling pathway. It acts as a receptor for TGFB1, TGFB2, and TGFB3, regulating diverse cellular processes. Mutations or dysregulation are implicated in several diseases, including Loeys-Dietz syndrome, Marfan syndrome, Osler–Weber–Rendu syndrome, and various cancers.
Ligand binding induces phosphorylation and activation of TGFBR1, which then propagates intracellular signaling cascades through Smad proteins or non-Smad pathways such as MAP kinases or PI3K/Akt pathways.
