Transforming protein RhoA (RhoA) is a member of the Ras superfamily of small GTPases and is encoded by the RHOA gene in humans. RhoA functions as a molecular switch, cycling between active (GTP-bound) and inactive (GDP-bound) states. It primarily regulates cytoskeletal dynamics, notably actin stress fiber and focal adhesion formation, by activating effectors such as ROCK1 and mDia1, thus controlling cell shape, motility, and contractility. RhoA signaling is crucial for immune cell activation and migration, cell cycle progression, and gene transcription, and is implicated in diverse pathologies including cancer, cardiovascular disorders, neurodegenerative diseases, and inflammation. RhoA is widely considered a key node in cell signaling and is a validated therapeutic target, particularly via inhibition of its downstream effectors such as ROCK (Rho-associated protein kinase).
