Transgelin-3 (TAGLN3) is a cytoskeletal protein within the calponin family, distinguished by a calponin homology (CH) domain, a single actin-binding site, and a C-terminal CLIK motif[3]. It consists of 199 amino acids and is highly conserved across vertebrates, with the human TAGLN3 gene located on chromosome 3q13.2[3]. Transgelin-3 is predominantly expressed in neural cells and plays roles in actin filament regulation, impacting neurite outgrowth, cell migration, neuronal differentiation, and possibly synapse formation[2][3]. It was originally discovered in the rat brain as NP25 and is mainly associated with the growth and differentiation of neuronal circuits. Although related transgelin/calponin family proteins have been implicated in cancer and inflammatory processes, there is currently no evidence that TAGLN3 itself serves as a therapeutic drug target or established biomarker. Its principal biological role appears to be in cytoskeletal organization, especially in neural tissues[2][3].
