Transient receptor potential A1 (TRPA1) is a nonselective cation channel and a member of the transient receptor potential (TRP) channel family. It is best known as a broadly tuned sensor for noxious chemicals, pain, cold, and itch. It plays key roles in pain detection, inflammatory responses, and airway inflammation. Many exogenous activators covalently modify cysteine residues on TRPA1’s N-terminal region. Endogenous activation can occur through signaling cascades involving phospholipase C pathways downstream of GPCRs like bradykinin receptors. Because it integrates multiple noxious signals leading to pain/inflammation, TRPA1 represents an attractive target for novel analgesics or anti-inflammatory drugs.
