Transient receptor potential canonical-5 (TRPC5) is a non-selective cation channel belonging to the TRPC subfamily of the larger transient receptor potential (TRP) superfamily. It is encoded by the TRPC5 gene in humans and functions as a polymodal sensor involved in various physiological processes, including neuronal signaling, cardiovascular regulation, sensory perception, and inflammation. TRPC5 forms functional channels as homotetramers or heterotetramers with other TRPC family members, especially TRPC1 and TRPC4. It is expressed in the central nervous system, endothelial cells, vascular smooth muscle cells, cardiomyocytes, sensory nerves, and fibroblast-like synoviocytes. TRPC5 acts as a receptor-operated non-selective cation channel permeable to Ca²⁺ and Na⁺ ions.
