Transient receptor potential canonical (TRPC) channels are a subfamily of the larger transient receptor potential (TRP) channel superfamily. TRPC channels are non-selective cation channels, primarily permeable to calcium (Ca^{2+}) and sodium (Na^+), and play crucial roles in cellular signaling by regulating membrane potential and intracellular calcium levels. The TRPC subfamily consists of seven members: TRPC1–TRPC7. In humans, six functional isoforms are expressed: TRPC1, TRPC3–TRPC7. TRPC2 is a pseudogene in humans and is not functionally expressed. These channels act as cellular sensors for various stimuli including mechanical stretch, temperature changes, osmotic pressure, pH shifts, reactive oxygen species (ROS), and G-protein-coupled receptor signaling. They mediate both receptor-operated calcium entry (ROCE) and store-operated calcium entry (SOCE).
