TRPA1 is a non-selective cation channel that functions as a sensor for pain, cold temperature, itch, and environmental irritants. It plays a central role in pain signaling pathways triggered by inflammation or reactive chemicals and is implicated in chronic pain, cough hypersensitivity syndromes, asthma exacerbations, itch disorders, and potentially hearing/balance dysfunctions. TRPA1 is considered a promising therapeutic target for analgesics or anti-inflammatory drugs.
Electrophilic irritants covalently modify cysteine residues within an intracellular "allosteric nexus" domain, inducing conformational changes that open the pore for cation influx. Calcium ions modulate potentiation/desensitization.
