Overview

The Transient Receptor Potential Cation Channel Subfamily C (TRPC) is a family of nonselective cation channels that play critical roles in cellular calcium signaling and are involved in various physiological and pathological processes. They are composed of six transmembrane domains, assemble as homo- or heterotetramers, and are activated by GPCRs and the phospholipase C pathway. TRPC channels are expressed in various tissues and contribute to cardiac contractility, vascular tone regulation, and renal function. Altered expression or mutations are implicated in cardiac hypertrophy/fibrosis, kidney disease, and pulmonary hypertension. Recent years have seen rapid development of selective small-molecule modulators targeting different subtypes for research and potential therapeutic use.
