Overview

TRPM7 is a bifunctional protein combining an ion channel with a serine/threonine kinase domain. It functions as a nonselective cation channel permeable to Mg²⁺, Zn²⁺, and Ca²⁺, regulating cellular uptake of these ions. It also possesses intrinsic kinase activity, phosphorylating itself and other substrates involved in cytoskeletal regulation and signaling pathways. TRPM7 is essential for embryonic development and plays a role in cell motility, adhesion, migration, polarization, proliferation, and survival. Aberrant TRPM7 expression is linked to cancer progression and metastasis, and it modulates platelet reactivity, affecting thrombosis and stroke risk.
