Overview

TRPM8 is a non-selective, voltage-dependent cation channel primarily known as the molecular sensor for cold temperatures and cooling agents like menthol. It is expressed in sensory neurons and other tissues, playing a role in thermosensation and potentially in cellular homeostasis through calcium signaling and regulation of autophagy. It is a target for analgesic drug development.

Mechanism of action

Activation by cold or chemical ligands (menthol, icilin) opens the channel, leading to Na+ and Ca2+ influx, depolarization, and action potential generation. Antagonists (AMTB) block this activation.
