Overview

Transient receptor potential cation channel subfamily M member 8 (TRPM8) is a non-selective, calcium-permeable ion channel that serves as the primary molecular sensor for cold temperatures and cooling agents like menthol (McKemy et al., 2002). It is predominantly expressed in small-diameter sensory neurons of the dorsal root and trigeminal ganglia, where it detects temperatures in the range of 10°C to 28°C (Peier et al., 2002). Beyond its physiological role in thermosensation and thermoregulation, TRPM8 is highly upregulated in prostate cancer tissues, making it a potential biomarker and therapeutic target for managing tumor progression (Henshall et al., 2001). In clinical pharmacology, TRPM8 antagonists are being investigated for the treatment of neuropathic pain, migraine, and cold allodynia, while agonists are explored for conditions like dry eye syndrome to stimulate tear production (Knowlton et al., 2013). However, a significant challenge in targeting TRPM8 is its role in systemic thermoregulation, as modulation can lead to adverse effects on core body temperature (Almeida et al., 2012).
