Overview

The **transient receptor potential vanilloid 1 receptor** (**TRPV1**) is a non-selective cation channel belonging to the transient receptor potential (TRP) family, specifically the vanilloid subfamily. It is best known as the molecular target for capsaicin—the active component in chili peppers—and is also referred to as the "capsaicin receptor" or "vanilloid receptor 1." Structurally, TRPV1 forms a tetrameric complex with each subunit containing six transmembrane domains; it shares architectural similarities with voltage-gated potassium channels. The primary biological functions of TRPV1 include detection of noxious heat (>43°C), mediation of inflammatory thermal hyperalgesia, regulation of body temperature, and transmission of painful stimuli from peripheral sensory neurons. It plays a significant role in various pathological conditions involving chronic pain and inflammation. Pharmacologically important ligands include agonists such as capsaicin and resiniferatoxin—which activate the channel—and antagonists like capsazepine that inhibit its function. Due to its central role in nociception and inflammation pathways, TRPV1 is considered an important therapeutic target for novel analgesics but presents safety challenges related primarily to thermoregulation.

Mechanism of action

Agonists activate the ion channel to allow influx of cations such as Ca²⁺ and Na⁺, leading to depolarization and neuronal activation. Antagonists block the ion conduction pore or allosteric sites to inhibit activation by endogenous or exogenous ligands.
