Overview

Transient receptor potential vanilloid 4 channel (TRPV4) is a non-selective, calcium-permeable cation channel of the transient receptor potential (TRP) ion channel superfamily, predominantly mediating sensitivity to osmotic, mechanical, and moderate thermal stimuli[1][2][3]. It is a homotetrameric membrane protein made up of six transmembrane domains per subunit, with large cytoplasmic N- and C-terminal regions featuring ankyrin repeat domains essential for protein interactions[1][2]. TRPV4 plays a key role in systemic osmotic pressure regulation, mechanotransduction, thermosensation, vascular tone, skeletal development, and epithelial barrier function in several organs including kidney, liver, lung, skin, and the nervous system[2][3]. Dysfunction or mutations in TRPV4 are associated with a spectrum of diseases, such as inherited neuropathies, bone dysplasias, and disorders characterized by abnormal fluid homeostasis or chronic pain[3].
