Overview

Transition protein 1 (TNP1) is a small, highly conserved, arginine and lysine-rich, sperm-specific nuclear protein expressed abundantly in elongating and condensing spermatids during spermatogenesis[1][2][3][4][5][6]. It operates as an intermediate replacement for histones, facilitating the transition to protamines, which enable highly compacted chromatin in mature sperm. TNP1’s main function is to relax nucleosomal DNA and promote histone eviction—key steps in chromatin remodeling—where it prepares chromatin for subsequent compaction by protamines[2][6]. Deficiency or dysregulation of TNP1 is linked to impaired sperm motility, abnormal morphology, increased DNA fragmentation, and ultimately, male infertility[1][3][6]. There are no known drugs targeting TNP1, and its primary relevance is as a molecular biomarker and mechanistic contributor in studies of male infertility.
