Transketolase is a thiamine diphosphate (ThDP) and divalent metal ion-dependent enzyme that plays a central role in the non-oxidative branch of the pentose phosphate pathway (PPP). It catalyzes reversible transfer reactions involving two-carbon units between ketose and aldose phosphates, thereby linking the PPP to glycolysis and enabling cells to adapt their metabolism according to energy and biosynthetic needs. Elevated transketolase expression has been observed across various cancers, supporting tumor growth. It is also a potential drug target against pathogens like *Mycobacterium tuberculosis*.
