Overview

Translocase of outer mitochondrial membrane 40 homolog pseudogene 4 (TOMM40P4) is a human pseudogene related to the TOMM40 locus. Unlike the TOMM40 gene, which encodes the channel-forming subunit of the mitochondrial import complex, TOMM40P4 is a non-coding RNA gene that does not produce a functional protein. TOMM40P4 shares high sequence homology with TOMM40, and its transcripts can be detected in human tissues; these RNAs may play a regulatory role by competing with TOMM40 mRNA for transcriptional or post-transcriptional protein interactions, or potentially generate small regulatory RNAs (such as siRNAs or miRNAs). However, there is no established biological function, druggability, or disease link for TOMM40P4. TOMM40P4 is not considered a therapeutic target, but its presence complicates molecular assays measuring TOMM40 expression, as conventional primers may co-amplify pseudogene transcripts[5][2][6]. Additional Notes: - TOMM40P4 should not be confused with TOMM40, which is an actual protein-coding mitochondrial transporter, associated with neurodegeneration risk, and considered a potential target in mitochondrial biology and Alzheimer's research[1][2][5]. - Literature does not support any unique or direct biological role for TOMM40P4 apart from possible post-transcriptional regulation[2][6]. - If TOMM40P4 appears as a therapeutic target in a research or screening context, this is likely a misannotation.
