Overview

Translocase of outer mitochondrial membrane 6 (TOMM6) is a small integral membrane protein that forms part of the TOM complex, the main entry channel for nuclear-encoded mitochondrial precursor proteins into the mitochondrion[3][6][10]. TOMM6 is an α-helical protein embedded in the outer mitochondrial membrane, closely associated with other core TOM components such as Tom40, Tom22, Tom7, and Tom5. Its primary biological role is to stabilize and modulate the higher-order assembly of the TOM complex, particularly in dimer and tetramer formations, which are required for efficient mitochondrial protein import[3][4]. TOMM6 is not a receptor, enzyme, or drug target in the classical therapeutic sense. Pathogenic roles have been tentatively noted via gene associations with developmental dysplasia of the hip and rare mitochondrial disorders, but functional and mechanistic links remain poorly understood[6][8]. No drugs are known to target TOMM6. TOMM6's alternative name "overexpressed breast tumor protein" arises from early findings of increased expression in some cancer tissue contexts, but it is not currently a characterized cancer biomarker or therapeutic target.
