Translocating chain-associated membrane protein 1 (TRAM1) is a multi-pass transmembrane protein localizing to the endoplasmic reticulum (ER) membrane and is a component of the protein translocation machinery. TRAM1 belongs to the TRAM/LAG1/CLN8 homology (TLC) domain protein family and is thought to play a supportive, rather than a receptor, role in the import of some precursor proteins into or across the ER membrane by influencing local lipid properties near the SEC61 channel but does not act as a selective substrate receptor[1][3]. TRAM1 associates closely with newly synthesized proteins early in their entry into or insertion into the ER and is required for efficient glycosylation and proper protein folding, as well as for ER-associated degradation during stress responses[1][3]. TRAM1-mediated processes are also implicated in certain genetic diseases and may influence viral immune evasion through ER-associated retrotranslocation mechanisms[3]. There are currently no known drugs that modulate TRAM1 directly, nor is it a typical therapeutic target such as a receptor, enzyme, transporter, or ion channel.
